MUTPRED The program MUTPRED generates mutability profiles of amino acid sequences from the corresponding cDNA sequence. The procedure is described in detail in Cooper and Krawczak (1990). The program generates UNWEIGHED profiles as described, and WEIGHED profiles for which the relative mutabilities are multiplied by the likelihood of clinical detection depending on chemical differences (Grantham, 1974). The program will run on all PCs or compatibles with a graphic adapter (VGA, EGA, CGA) installed. If the program fails to run on your computer, you may try to run the program EMUCGA.COM in advance. MUTPRED does not produce any hardcopies. To get a hardcopy, use the PRINT SCREEN key on your computer. Alternatively, the ave option allows one to download the mutabilities actually displayed on the screen in a user defined file. You may then use your own graphic software to transform these data into a profile. MUTPRED is initialized by the file MUTPRED.MSG which has to be installed in the current directory. After the program has been initialized successfully, MUTPRED asks for the name of the cDNA sequence file. This file should contain the plain sequence information in the correct reading frame, including nothing else than spaces and the four characters A, C, T and G. You may include comment lines starting with a semicolon ';'. After a few moments, the profile appears on the screen. Strike the '+' or '-' keys to scroll the profile, 'u' or 'w' to generate unweighed or weighed profiles, respectively. Strike 'f' to load a new file, 's' to download data, 'e' to finish. Best wishes MK
